Polymericmicelles,self-assembledsupramolecularstructuresofblockcopolymers,arepromisingnanocarriersystemsfordrugandgenedelivery1,2).Severalmicellarformulationsofantitumordrugshavebeenintensivelystudiedinpreclinicalandclinicaltrials,andtheirutilityhasbeendemonstrated.Evencomparedwithlong-circulatingliposomes,polymericmicellesmighthaveseveraladvantages,suchascontrolleddrugrelease,tissuepenetratingabilityandreducedtoxicitysuchashand–footsyndromeandhypersensitivityreaction.Importantly,criticalfeaturesofthepolymericmicellesasdrugcarriers,includingparticlesize,stability,andloadingcapacityandreleasekineticsofdrugs,canbemodulatedbythestructuresandphysicochemicalpropertiesoftheconstituentblockcopolymers.Thedevelopmentofsmartpolymericmicellesthatdynamicallychangetheirpropertiesduetosensitivitytochemicalorphysicalstimuliisthemostpromisingtrend,directingtothetargetingtherapywithhighefficacyandensuredsafety.
